Bio-FabLab Laboratory Intro

Friday the 22nd of March we participated in Bio-FabLab’s Laboratory Introduction Seminar.

We learned how to handle pipettes, Erlenmeyer flasks, Bunsen burners and a lot of other laboratory equipment. We were also instructed in safety procedures and learned how to work in and create sterile environments for handling bacteria ect.

The main lesson of the day, was instructions on how to create our own Agar plates, used for growing bacteria. Below are some of the steps and practices we did and learned during the seminar.

In our project, we will need both Broth and Agar plates to handle and cultivate our bacteria, in the list of ingredients the difference between the two is shown. Agar makes the solution solidify, and the solution is poured hot into petri disks.


List of Ingredients:

Tomato Broth:
- 25mL Peptonized Milk
- 100mL Tomato Juice
- 125mL Distilled water

Tomato Agar Media:
- Add 5g of agar to the solution.

Agar, TSB and Sand Prep.

Tuesday the 9th of April we created our first Urea modified Agar plates. We also made our Triptic Soy Broth (TSB) and experimented with Autoclaving of sand.

Our TSB and Agar plates needs to be modified with Urea, else the bacteria will die, and experiments fail. The Agar plates gets directly infused with Urea, but the TSB will be mixed with Urea every time we use it, as the Urea will undergo a natural process and become less potent.

List of Ingredients:

TSB 1L (2L is needed):

-   Destilled Water         -   0,9L
-   Tryptone                -   17,0g
-   Soytone                 -   3,0g
-   Sodium Chloride         -   5,0g
    [NaCl]
-   Dipotassium Phosphate   -   2,5g
    [K2HPO4]
-   D-glukose               -   2,5g

Added to the TSB solution In smaller doses when TSB is used:

-   Destilled water         -   100mL
-   Urea                    -   20,0g

Urea modified Agar Media 1L:

-   Destilled water         -   1L
-   Tryptone                -   10,0g
-   Yeast/gær               -   5,0g
-   NaCl                    -   10,0g
-   Agar                    -   20,0g
-   Urea                    -   20,0g

1M NaOH and/or HCI (to adjust PH LVL’s)

First Streaking

Wednesday the 10th of April we used our newly created Agar plates to streak our bacteria solution for the very first time. We streak our bacteria in order to isolate single colonies for future culturing.
